Production Process of Penicillin Solid Preparations
 

Strain fermentation

• Inoculate Penicillium flavum producing fungi on solid medium and incubate at 25°C for 7-10 days to obtain Penicillium spore culture.
• Inoculate the spores into the sterilised medium in the seed vessel by making a suspension of the spores with sterile water, passing through sterile air, stirring and incubating at 27°C for 24-28 hours.
• Inoculate the seed culture into the sterilised medium containing phenylacetic acid precursor in the fermentation tank, pass through sterile air, stir and incubate at 27°C for 7 days. During fermentation it is necessary to replenish the phenylacetic acid precursor and the appropriate amount of medium.

Extraction and purification

• The penicillin fermentation broth was cooled and filtered. The filtrate was subjected to multi-stage countercurrent extraction with butyl acetate in an extractor at pH 2-2.5 to obtain the butyl ester extract.
• The butyl ester extract was transferred to a buffer solution of pH 7.0-7.2 and then to butyl ester, decoloured with activated carbon, added with a salt-forming agent and distilled azeotropically to give penicillin G potassium salt.
• Penicillin G sodium salt is prepared by passing penicillin G potassium salt through an ion exchange resin (sodium type).

Solid preparation

• Penicillin G potassium or sodium salt obtained by extraction is dried to give penicillin solid powder.
• The penicillin solid powder can be further processed into solid dosage forms such as tablets and capsules as required. This usually involves process steps such as tablet pressing, filling and coating.
