The most commonly used method for preparing suppositories is the hot melt method.
The process flow of preparing suppositories by hot melt method is as follows: melting the matrix, adding drugs (mixed), injection molding, cooling, scraping, and taking them out.
First, wash and dry the bolt mold, and apply a small amount of lubricant to the inside of the model. Then add the medication in different ways according to its properties, mix well, pour it into the suppository mold until it slightly overflows the mold opening, let it cool, wait for complete coagulation, cut off the overflowing part with a knife, open the model, and push out the suppository. This method is applicable for the preparation of suppositories with fatty and water-soluble matrices.
(1) The method of adding suppository drugs: ① Insoluble drugs should generally be crushed into fine powder and mixed well with the matrix;
② Oil soluble drugs can be directly dissolved in a melted oily matrix;
③ Water soluble drugs can be directly mixed with a melted water-soluble matrix; Alternatively, absorb an appropriate amount of lanolin and mix well with an oily matrix.
(2) Lubricants:
The hole of the suppository mold needs to be lubricated with lubricant to facilitate the removal of the suppository after condensation. There are two commonly used types:
① The suppository for oily matrix is commonly made into an alcohol solution consisting of 1 part soap, 1 part glycerol, and 5 parts 90% ethanol.
② Oil based lubricants such as liquid paraffin and vegetable oil are commonly used in suppositories with water-soluble or hydrophilic matrices.
